Brandon Richard Flowers nació el 21 de junio de 1981 en Henderson, Nevada, una pequeña ciudad cerca de Las Vegas. Desde temprana edad, Brandon mostró un interés marcado por la música, influenciado principalmente por la colección de vinilos de su hermano mayor y por la vibrante escena musical de Las Vegas. Su familia, de raíces mormonas, se trasladó a Utah cuando él era adolescente, pero Flowers pronto regresó a Las Vegas, la ciudad que moldearía su carrera musical.
En 2001, después de asistir a varios conciertos locales, Brandon respondió a un anuncio en una revista que buscaba músicos para una nueva banda. Allí conoció al guitarrista Dave Keuning, con quien formó la banda que más tarde se conocería como The Killers.
